just made my own estrogen vials using the powder i bought 2 years ago. unfortunately i didn't have much powder to experiment. but estradiol enanthate does dissolve completely at 60 mg/mL with benzyl benzoate 40% and benzyl alcohol 3%. tomorrow i'm going to sterilize them with instant pot. unfortunately the cheapest model only has 11.6 psi so i had to pay a little more for 15 psi.
next time i'll buy undecylate to make 100 mg/mL.

estrone and estriol have short half-life and low affinity for estrogen receptors
unless there's an effect on LH suppression or attenuation of SHBG increase, these are just fairy tales dr powers uses to waste his patients time while an immense improvement in healthcare might be made with advocacy for oral estradiol undecylate capsules. there's a precedence with testosterone undecylate capsules, there are studies from the past century which prove it works and avoids first pass effect. why wouldn't he design a clinical trial with measurement of coagulation factors etc. and campaign for such capsules, not only for trannies but also cis women?

i prefer injections too, but i think about people who fear injections or can only access pills, sublingual and gel. undecylate would change everything for them, it would be so much more convenient while preserving benefits of parenteral estrogen.

>so basically its like injections but in pills and no e1?
capsules, not pills. estradiol undecylate is dissolved in MCT oil and transported in a capsule to avoid first pass effect. since undecylate is an extremely long ester, it allows to preserve half-life long enough to dose it daily.

With EUn injections it's typically dosed once a month, which is even easier.
Injections are the best route of administration in almost every way, and they always will be.

This is my first post here so sorry if my questions are stupid or medical wording is wrong, but I have a few to ask. any help is very appreciated <3

One; is there any truth to the idea that it's better to start on oral hrt (before switching to injections around 1yr) because it produces lower but more consistent levels & liver pass metabolites more closely resembling the early stages of cis female puberty? My only trans friend is convinced of this, did it herself, and she had great tits. I started at 4mg of oral and take 8mg now (along with 100mg of spiro) and am very surprised how much growth I've had so quickly. i've had people ask me "why don't you take it sublingually?" and im honestly not sure if that's better? but im worried about changing anything when it seems to be going about as well as it can right now. so what's the reality of it?

Two; I really want to switch off spiro (costly dehydrated, peeing a lot, light headed & the color leaves the corner of my vision if I get up too fast) but my doc only offered dutasteride or finasteride as alternatives (ive never heard of those as main AAs??). She said "your insurance likely won't cover bica" and I didn't push it. So I'm considering getting bica DIY (fuck spiro + bica supposedly promotes breat growth) but am concerned it'll make my t level drastically shoot up. Ik it won't cause masculinization because of how bica works but my doc will notice if my t shoots up on bloodwork. Are there other realistic options to DIY as an AA in the USA? I've heard cypro causes tumors and the only affordable GnRH agonist i've found is a nasal spray every 8 hours (buserelin) and that it can potentially cause diabetes. what is the truth & what is my best option?

If you really want a GnRH buy relugolix from OpenGateLabs—it's an antagonist rather than an agonist, but it works just as well and is quite affordable. Plus they ship US domestic.

ask your doctor for depo-provera. it's once 3 months contraceptive - medroxyprogesterone, works like cypro. based on studies on men it's capable of completely suppressing testosterone on its own. idk how it is in your country but in mine depo-provera is only 13 $ per injection.
anyway, if your testosterone is within female range on e+spiro, it won't increase after discontinuation because spiro is only an androgen receptor antagonist in clinically used doses.

monotherapy is able to completely suppress gonadal testosterone with a high enough dose ALWAYS provided you don't have a tumor or EXTREMELY rare estrogen insensitivity (whose symptom is still growing taller way after puberty). estrogen suppresses gonadotropins. gonadotropins signal to Leydig cells in testicles to secrete androgens. it's only a matter of how high dose you need, but with 400 pg/mL (still safe) estrogen testosterone won't be suppressed only if it wouldn't be suppressed by cypro or GnRH agonists anyway.

could someone please give me a hand estimating what dose i should be on? I'm a bit lost desu

I've been on hrt since 19/02/2025 and I've gotten my most recent blood test back and I'm a bit unsure of how much to increase it by. I've obviously been hondosing myself but for my previous test done just after 3 months i was massively overdosing myself so I'm kinda wary of doing that again.

anyway here's my results after injecting 4mg EE from astrovials every 7 days for 2 months

>Estradiol - 431 pmol/l
>FSH - <0.30 U/l
>Free Androgen Index - 7.75
>Luteinising Hormone - 5 U/l
>Progesterone - 0.83 nmol/l
>Prolactin - 476 mlU/l
>SHBG - 28.35 nmol/l
>Testosterone - 2.196 nmol/l

So obviously my T is a bit too high and my estrogen is a bit too low but I think I'm closer to my target ranges than my previous test, which I'll post below

>Progesterone - 0.27 nmol/l
>Prolactin - 512 mlU/l
>SHBG - 170.9 nmol/l
>Testosterone - 0.21 nmol/l

so yeah obviously everything was much better supressed here but the shbg and estrogen were so ridiculously high it was basically useless anyway. this was after 2 1/2 months of 5mg EV every 5 days and 2 weeks of 5 mg EE every 7 days. All SubQ injections btw

bullshit. shbg is irrelevant for estrogen bioavailability. it only makes the gains less proportional, but the gains still happen. LH wouldn't be completely suppressed if there were less estrogen action, inhibition of LH is mediated via estrogen receptor alpha which is responsible for practically all feminization.
i fucking hate you for using trash molar units. you changed the vial which makes changing dosing more tiresome. i guess that it's estradiol enanthate 50 mg/ml without benzyl benzoate and it causes incomplete dissolution, decreasing absorption. or there was a problem with filtering. or you miscalculate the amount of fluid to draw.

Sup legbutts. I'm injecting 0.40ml (vial with 40mg/ml) of estradiol enanthate every 14 days. Whenever I get to the last day before the next shot, my boobs feel more sensitive and my face gets way less oily. Does that mean I'm injecting too much and my levels only feel "normal" at the end? Should I lower the dose or just keep it as it is?

Can high estrogen levels cause edema? For the past couple of months i've been getting a bit of swelling in my ankles after sitting on my ass all day at my dumbass office job. This coincided with accidentally dosing myself too high after switching to EUn, my levels were 350pg/mL at through. Thing is i already did my shot so i'm stuck like this for another month, it's not like i can adjust my dose super quickly with undecylate and see if the edema fixes itself.

Also i need some help interpreting LH, FSH and SHBG. I could not find any info on what they should actually be in trans women.
My SHBG is 91 nmol/L, the reference male range the clinic gave me is 11-54. Apparently 91 it's within range for cis women, does that look normal given my high E2 levels of 350 pg/mL?

My LH is 0.61 mlU/ml (reference range 1.24 - 8.62) and my FSH is 0.37 mlU/ml (reference range 1.27 - 19.26). These seem nuked enough given my T is clearly suppressed at 0.13 ng/mL, right?

estrogen ok, testosterone is measured with mass spectrometry meaning very accurate measurement. 53 ng/dL with such high estrogen is concerning about adrenal androgens (you're supposed to have no more than ~15 ng/dL in such conditions). if you have any acne, you'll benefit from low dose bicalutamide.

The most bog standard dose and ester is 4mg EEn (enanthate) injected once a week. Cypionate has a very similar half life curve so you can alternatively use that. It's a well studied ester that is largely consistent between people and will allow you to adjust your dose quickly, unlike EUn (undecylate) which has a much longer half life (you can inject monthly), but is less understood and there is a lot of trial and error to fine tune your dose, and it takes months for your levels to stabilize before you can get tested. Valerate is also widely used and understood, but it has a very short half life and leads to less stable levels.

You need to calculate the actual volume in the syringe based on the vial's concentration, for example for a 50mg/ml vial, 4mg would be a 0.08ml dose, or 8 units in an insulin syringe.
Speaking of, buy small insulin syringes and do subcutaneous (subQ) injections, no need to muck around with changing needles or deadspace.

You pick a spot to inject on the region of your thigh in picrel. You just disinfect the injection spot and the top of the vial, you draw the same amount of air into the syringe as you are going to draw from the vial (8 units), you insert the needle into the vial straight down, inject the air, flip the vial upside down then draw that amount of liquid out. Don't worry if there are small air bubbles left, they are completely harmless for IM and subQ injections. Then stab yourself with the needle, not too violently but also in a swift motion without hesitation. Raise the plunger for a few seconds to see if you draw any blood so you don't inject in a vein - if you do just take it out and insert it again a couple of centimeters away, no big deal. Inject the dose, wait 10 seconds then take the needle out. I personally press down on the injection spot for a minute with an alcohol wipe to prevent anything from leaking out.
Don't forget to switch thighs and alternate injection sites between each injection.

Also in most countries you can just order insulin needles online, or walk into any pharmacy and say they're for your grandma or whatever. Get blood tests at through (right before your next dose) every 3 months and adjust your dose as needed so that your E is above 100 pg/ml and your T is suppressed.

I'm not aware of any pathways for E to be converted to DHT. Are you on prog? That can get converted to DHT. You sure you weren't balding beforehand? E won't make you go bald but it might make your hair thinner, which would make existent balding more apparent. Ultimately you'll have to get a blood test to see what your T, DHT and E levels are to figure out the cause.

No, not unless you're injecting almost parallel to the skin

there is a chance, it's happened to me before. that's why you aspirate before you inject. the amount of oil you'd inject isn't dangerous even if you were to somehow inject your entire dose into a vein anyway, but the dose would be ineffective

4mg a week is enough? does 4mg daily sublingual just get absorbed a lot worse?

They're just not comparable, 4mg of pills are not the same as 4mg of gel or injections or patches. Even the same pills will get absorbed very differently based on whether they are taken orally or sublingually, and gel will be absorbed worse or better based on the area of the body it is applied to for example. Different administration methods with different ways of entering the body will each have unique absorption. Not even different injection or pill esters are completely comparable between each other, the same dose of valerate and enanthate injections will give you different levels for example even disregarding the half life (see image), and also 2mg of EV pills is equivalent to about 1.5mg hemihydrate.

4mg is a pretty standard EV dose and should put most people at over 200pg/ml at trough and lead to adequate T suppression.
You can use this calculator to play around with different doses https://transfemscience.org/misc/injectable-e2-simulator/

EEn*
Also keep in mind that injections will create a depot site which is slowly absorbed into the blood stream over the course of weeks. When taking pills sublingually they will basically enter the blood stream almost directly and get eliminated quite quickly which is why you need to take them multiple times a day. Swallowing pills will make them enter the blood stream slower through the digestive system, but the estradiol will be processed by the liver and a lot of it will be destroyed/turned into estrone, which is why that leads to lower levels than sublingual.

not really, i didn't feel anything different. there was just a tiny droplet of blood at the base of the syringe when i inserted it, then when i pulled back the plunger a little more came through. i actually took a photo of it because it was so unusual. when that happens you can just insert the syringe in a different spot, the tip shouldn't blunt too much from just one insertion.
