What would be a threat big enough to make Twilight go to the princesses to request that they give her their alicorn magic? She would be so desperate that she would have to take it from them by force if she had to. Draining their bodies to the point of near death just to gain the power she needs to defeat the threat at large. Let's also assume her friends are out of the equation. They either got killed, captured, or worse.

I'd say Daybreaker could be a sufficient threat. Or Midnight Sparkle. Or Anon, the human, coming through the portal and being mostly immune to magic and owning a gun with lead that's from earth and thus immune. Grogar is also maybe strong enough. Trixie getting the memory stone could also be a problem. Pinkie Pie could be a problem, considering her power level. Terik, also, was always a Chad. The sea ponies or a federation of different species challenging ponies for supremacy. Discord, obviously, can be a problem. Sombra is Pony hitler. Capitalistic nobility could be trying to revolt against the monarchy.
