What ever happened to stem cells?

20 years ago everyone was up in arms they were not allowed to have abortions to make stem cells and the stem cells were going to cure all ailments

whta happened?

A few points, but not exhaustive: Stem cells proliferate and differentiate into different cell types. The proliferation of said cells is easy and desirable during development, but in an adult organism the same behavior requires the activation of genes cancers also use to proliferate. Even if the stem cells don't become cancerous the immune system can target said cells as though they were cancer cells. You need a way to control a stem cell's activity so it proliferates enough to replace damaged tissue without overproliferation and ideally without suppressing the immune system.

Other problems present for getting stem cells to the appropriate locations and acting as another cell of the same cell type. For example, if you want to regrow a neuron in a parkinsons patient you need to get a candidate cell to differentiate into a neuron, get the replacement neuron where it's needed, and recapitulate the connections the previous neuron had to other neurons in the new neuron. There are a litany of problems doing all this.

In short, stem cell therapy is very complicated (though I wouldn't say it's impossible).

Transplantable organs specifically also have unique challenges. Last I heard we didn't have sufficient ways to deliver oxygen through blood vessels grown within organoids to make them fully sized and functional organs. In the short-term cryonics and removing endogenous viruses from organs of close-enough species to us are decent candidates, but again have a lot of problems that aren't fully solved.

Depends. I think we'll see life extension as a result of bioengineering this century, but not biological immortality or eternal youth. If we get AGI and government resources align around biological immortality maybe but I don't have knowledge of either to comment on them.

I think if eternal youth can happen, it will be within the next few hundred years. Maybe we're in the first generations of people who could theoretically make it to that point (whether by preservation or alignment of biotech and policy) but I doubt it. Still going to work towards it, though.

This tech is now used in hospitals and the initial drama is over. One of my neighbours survived cancer thanks to stem cell treatment with stem cells from a relative.

It’s possible to exhaust stem cells and I’m not sure they have enough research into what keeps them continuing. That’s a question. So there’s lots of areas of study to support stem cells.

Inhibiting prolyl hydroxylase domain inhibitors will keep the growth going? The most significant source is in store bought taco shells. And growth factors. This is the opposite of what is known as “healthy.” Because it increases fibrosis and probably triggers remodeling. I don’t know what method cleans up the area and remodels back to normal instead of a bad adaptation around an injury or old tissue.

I suppose the correct way to fix an injury;
1. Appropriate blood flow and pressure
2. Waste & fibrotic tissue removal
3. Stem cell proliferation
4. Regrowth / new elastin/collagen

The block to stem cells is the old shitty cells, abnormal blood flow, and waste/plaques/damage/old cells/cancer/bad or excess collagen.
The problem is also the localization vs systemic, where you might not want the drugs going everyplace. That’s why they use hydrogels. Obviously it’s not a casual thing to open yourself and and apply a hydrogel. You can make external ones for skin though. You could mix drugs or stem cells into store bought gels maybe.

It’s a super wide research area with overlapping into other areas, like even with exosome / secretome and cell signaling research.

this thread still up? i didn't see the correct answer so i'll provide it now. Mesenchymal Stem/Signalling Cells derive their usefulness not from differentiation but by mediating inflammation/healing via exosomes. and to your point, most if not all of the innovation has continued offshore ever since bush's moratorium. even the efficiacy of MSCs are muted if limited by autologous same-day cell expansion as currently allowed by us laws. included is a talk from michael west concerning stem cells.
