I was there in the early fandom. Plenty of first watchers were asking if Dash was female. While having eyelashes is a potentially big tell there are two things:
1. It's a cutsey show, and initial perception of this show could absolutely allow the idea that males also have eyelashes in this.
2. Her eyelashes are straight and thin and has only 3, while other mane ponies have thicker, more numerous eyelashes (except AJ).
Also it was not like it was a long-term perception a lot of poeople held. Just first knee-jerk reaction. Sooner or later in the episode they happen to watch first some pony drops a "she" or "her" when referring to RD and the uncertainty is gone.
One more strange thought of this kind I remember from watching episode 1 was: "can stallions speak?". It seemed weird that guards that delivered Twi to Ponyvile only made horse noises. Of course it was gone shortly after.
