Put your mouse over the pixel you want to mute with (pic related) and press F1 when it's the color you want it to be when it should not be muted. If it deviates from that original color it will mute the video. You can select multiple pixels, just go over the pixel you want and press F1 again. Press F2 to clear all selected pixels.
Weaker hardware can drop a lot of frames from the constant pseudo-screenshots. Tweak the poll_time if it's too much. I've got it running on my 10 year old HTPC muting the donation reader and it's totally tolerable though.
There is a some lag in the AGDQ visual cue, adding "mpv --audio-delay=0.1" can help auto-mute before hearing someone speak.

Have the video player mpv.
Download the file as auto-mute.lua and put it in your mpv script directory "~/.config/mpv/scripts/" on linux/macos or "C:\users\USERNAME\AppData\Roaming\mpv\scripts\" on windows, make the directory if it doesn't exist yet.
Watch the stream or video and use F1/F2 as described.
Alternatively command line something like
mpv --script=~/Downloads/auto-mute.lua https://twitch.tv/gamesdonequick

Yes. Some will use it and some won't. I think it is best to put power in the hands of the viewer to have more control over their experience. You can auto-mute VODs as well.

Not really. All the audio is in one stream so it's not possible to mute the voices and keep the game audio. This is a trade-off I am willing to accept.
