The "Red Line" was created using one of the ancient weapons or in conjunction with it. The huge dense mass suddenly appeared in the ocean, causing sea levels to rise. This split the seas up and separated them away from each other.

Luffy is gonna fulfill the prophecy and destroy the red line, uniting the seas/world/map back into "One Piece"!

The Mural is referring to the four seas, not anyone in particular. It references the oceans meeting together as one!

I can't believe I never realized it. The Red Line!!!! A massive unnatural structure that appeared in the ocean and seperated the sea, splitting it up, The Great Serpant is the Red Line!
