In Curse of Darkness, the effeminate dude is Isaac, who isn't even a vampire, and wants to murder Hector with passion because he blames Hector's defection as betrayal and contributing to Dracula's death in the preceding game. But he goads Hector because the soul of a Devil Forgemaster can be used to revive Dracula.
While the bishounen designs started getting used in the 3D games and later entries, they were still as straight as they could get.
