Best designed is a nebulous term. However, going by common criteria, we can identify several key factors; confluence of design, versatility of design, and actual usefulness of design.
Confluence is in regards to how every element interacts with each other. Ideally, each element supports the other, whether it be synergizing for greater effect or covering for the weaknesses of other elements.
Versatility is just that, the ability to do many things with one function. When an operator can fulfill multiple purposes whether it be from utilizing their different skills or their skills themselves being multi-purpose, it is advantageous for Rhodes Island and shows complex/considerate design.
Last, but very far from least, usefulness. The other 2 factors become non-factors if the design is not functional in the first place. Being useful is the inherent criteria to all mechanical design in existence. However, what is useful is in the eye of the beholder as well as the ability to reject usefulness as a criteria altogether and see beauty in design for other reasons.
For an example of an operator I would appraise well in all 3 categories, operator Saria qualifies highly. As a Guardian, she has abilities synergistic for healing, herself and others, supporting, through SP recovery and debuffing, while being able to hold the line herself. Her talents aid all her skills and all her skills have their own use cases. Each of which have all seen their applications during her tenure on Rhodes Island.
To give a counter-example that does poorly across the 3 categories, I would unfortunately present operator Fartooth.
