remember, in 2 the first murder was an 'accident' in the sense that the killer didn't want to kill the victim, and the victim sacrificed their life to save another, the killer also deluded themselves into thinking that killing their intended target would protect everyone
this somewhat made everyone take the situation less seriously than if it was a flat out cold blooded murder, they sorta took nagito seriously, but that was the extent of it
if it were a deliberate murder of the intended target for the purpose of escaping, i see everyone taking things much more seriously and not willing to put themselves into situations where they could get axed
what i'm trying to say is i don't see fuyuhiko killing mahiru in the second case because i don't think she would allow herself to be put in that situation if case 1 didn't happen the way it did

Despair disease if that's still a thing. Nekomaru on despair disease could be a good 'horror' moment for the game, to have the loudiest and wackiest guy in the game to just get really quiet and on edge during a trial.
But in the main game, him and Gundham agreed to fight to save everyone essentially. Whoever committed the murder would they have to play into the class trial, and it was done to ensure they didn't die in the funhouse, so something like that could also happen.

But Despair Disease in general makes it so that 2-3 could have ANYONE be the killer.

The difference here is that it's still telling you what is happening. Is it being vague for the purpose of hiding that she's planning a murder? Yes. Is it omitting her actions? No. If that line instead just said something along the lines of "I ran after Shuichi" and didn't mention that she dropped something I would have a bigger problem with it. Meanwhile in order for the chapter 6 prologue to be the real one there is zero indication that there is more happening in the scene than what is being shown and you're omitting dialogue.

I think Chiaki is definitely going to die first since Monomi's accomplice dying would make the scenario the most different out of all individual deaths. Then it would make the most sense to me that her role as the lead support girl gets overtaken by either Mahiru, Or Peko if Fuyuhiko dies in the second case.
Also they are definitely going to use the impostor for something this time around. Maybe using the Nekomaru disappearance event by with fake Byakua killing him and hiding the corpse for over one trial, while impersonating Nekomaru. Though there would have to be a plausible reason for the fake Byakua to not show up on the trial.

2x2 might retcon dr3 but not because it will be a sequel to it. In the store page it is stated that "A new killing game begins in the same setting and with the same characters as Danganronpa 2: Goodbye Despair". That to me heavily implies that this will act as an alternatative scenario to dr2 specifically (Since the same setting will still be the simulation), which might in turn cause the events after it to change If the cast of dr2 is unable to escape or something different happens within the confines of the killing game. I'm not sure how else people would interpret "same setting" unless there is a hidden remnant of Junko's influence somewhere in the world post dr3 which we are unaware of, or it ties into v3 somehow. However, it's not impossible for the new game to tie into dr3 even if a lot of the story changes since the events that happen within the killing game of dr2 are more or less inconsequential as long as Izuru wakes up from it. They can basically just rewrite it with different happenings as long as it can logically tie to that same conclusion. Now if I had to take an educated guess, it would be that this won't be tying to the main timeline. Why? Well it's simple really, that would be quite boring. Since we now know that the murders that happen in the simulation setting(except from Hajime for reasons stated above) have no consequences in the real world it brings down the weight of the events a lot compared to something like dr1 and v3. It only makes sense for the sake of a more compelling story, that the writers either have this new "drastic event" significantly alter the plotline as we knew it in dr2 or that they tie it with an alternate universe (either v3 or an entire new concept that would change the story before the killing game).
