Either he shortened his lifespan massively by abusing his instant regeneration, dying of dividing his cells until they ran out of telemeres, or he died of cancer and that cancer is what they call Hashirama cells.
In real life there's an ancient dog that mutated to become a transmissible cancer, and then there's human cancer strains that go on forever if they're cultured for research purposes. Hashirama cells going out of control and expanding into vegetatation biomass is very tumor-like, as we saw with Danzo and the defeated white zetsu.
Hashirama might have just exploded into that huge tree in the village that gets uprooted during the 9 tails attack.

Edo tensei revives you in the state you were in when you died, with all the weird particularities that entails. Nagato's prime would have been when he was a healthy teenager but he gets revived as a shriveled, debilitated husk with his hair color drained from the moment he used rinne rebirth.
The only exception is Madara who Kabuto presumably utilizes dna from two periods of Madara's life to fenagle him a unique edo body that can alternate powers.
