Uro said that she was "probably" in love with Madoka but that the relationship between the two wasn't sexual in nature. Since this interview he's re-iterated that their relationship is "love-like". Homura's VA has said similar things as well but i can't find the interview for that right now. I can't canonically have sex with her since she isn't real and i don't exist in the universe of Madoka Magica, but that doesn't change the fact that i want to have sex with her and have in my dreams multiple times.
I do think Homura as a character works better if she is a dyke for Madoka, but in the end she officially isn't one according to Urobuchi, even if she does behave like one. I do hope this gets retconned during Kaiten, mostly since i think the meltdown that this would cause in these threads would be funny, and because the aforementioned fact i think Dykemura works as a character.

Homura being motivated by romantic love for Madoka also doesn't make much sense. The plot would be fundamentally different if she was. She obviously wouldn't be trying to get Madoka as far away from herself as possible and save her from being a magical girl where she would be with Homura all day. By trying to prevent Madoka from contracting, Homura is trying to create a situation where she and Madoka will essentially not belong to the same world, and will spend much less time together than if Madoka was a magical girl. Of course if she was gay for Madoka she would actively be trying to spend as much time as possible with her, and them being magical girls together would help that.
