Probably not with Matt and Trey but the individual animators and storyboarders who worked on other things most definitely dealt with or met Kyle at some point. He was very active in those communities for years. The similarities between him and the pic are a bit too close to ignore as a totally random event. Seems like he creeped out someone at some point and they wanted to make fun of him later on while working on South Park.

Why bother? They are all incredibly simply cookie cutter simplistic concepts. They're all watered down Cow & Chicken/Ren & Stimpy/Schnookums & Meat in some form. Two buddies who happen to be some weird pun named things like two animals, or objects having wacky wild adventures. It's the same thing overwhelming majority of the What A Cartoon shorts happened to be and never went anywhere back in 1995.

What is stunning is how Kyle never could figure out that no one had been making a show like that in years, and they really had no audience any longer. That type of thing was only successful a couple times, a failure lots more times, and died out in the mid 90s.

It was kind of working over at Nickelodeon when they did Sanjay and Craig, and then Pig Goat Banana Cricket, and the one with the talking cheese, I forget the others. There were a few of those poorly named buddy wacky toons happening. But they all followed a much simpler style. Maybe if Kyle changed up the look to match what they were Greenlighting (The advice almost anyone working in a studio would tells everyone all the time!) he might have gotten a chance.

Disney tried Pickle and Peanut but that also failed. But those shows were still sorta happening.
