Well, honestly. I wouldn't change a whole lot so much as I would just start putting character's on a bus/having them fade into the background. There's more than I can probably remember off the top of my head, but for example: Charlotte, Lucy, Catalina, Rhoda, Ashley, Jay, Liz, Justin, George, Tensaided, Greg, Noah all could get sidelined and not much would really change/no one would really care.
If I were to make any big changes, I think it would be having Justin sort of phase out of the main cast after the birthday party/when Elliot and Susan begin their review show. Ever since the birthday party he hasn't truly been a main cast member or had anything interesting happen with him that I can recall. He just sort of exists to give the characters a reason to hang around the comic shop for slice of life stuff. The only time he's done something interesting was when he kicked the fire guy and that was after a long boring period and was followed by an even longer more boring period that still hasn't ended. Anything you wanted to do with him could easily have been replaced by having Justin and/or Susan hanging out at work talking with adding in George or Tensaided as needed.
I'd also probably combine George and Tensaided into one character since they fulfill basically the same role in the story and neiter are that important. In fact, if I could get away with it I might just replace them both with Sensei Greg and just have the Dojo be a back room of the comic shop/video store and have the joke be that it's what actually pays the bills (it would also work thematically with the Anime Style Martial Arts Dojo.)
I would probably have the video store and comic shop be the same place like a type of Sam Goody since there's really no need for them to be different places at all narrative-wise.
