Isekai is a genre of anime about people being sent to/trapped in fantastical other worlds, usually fantasy themed, usually as a power fantasy. Normal every day guy gets dropped into a fantasy world, learns magic, becomes a badass, gets a harem of cute girls, etc. Reincarnated as a Slime, Mushoku Tensei, Konsoba, etc.
"Trapped in a Video Game" isekai is a subgenre of that, where the world they are in is explicitly a video game, and runs on video game mechanics. Log Horizon, Sword Art Online, etc.
Villainess Isekai is a subgenre of even that subgenre, that is focused specifically on otome games. Basically dating sim games aimed at women, there the player plays as some bland but pretty girl having to navigate conversations with pretty boys and win their love. A stereotype of this genre of game is the "villianess" character, who is a female antagonist that exists to be a huge bitch to you and fuck your shit up.

Villianess Isekai is the specific and surprisingly large subgenre of characters getting trapped in a video game, but SPECIFICALLY getting trapped in the game not as the player character, but as the villianess character in these games. Who is, by definition, an important and mean female character... but also a character thats fated to LOSE in any proper ending of the game.
So the stock setup here is that a person who is very much not suited to the role of villainous (a meek/nice girl, or any kind of dude) being stuck as a villainess and having to figure out how to derail a game they are designed to lose so they don't end of exiled or beheaded or whatever happens to them in the normal game, in a game whose rules all hinge around dating cute boys.
