You're talking about tens of thousands if not hundreds or millions of very very specific compounds and combinations to test. Every minor tweak to the mixtures and what molecules are bonded to what and where can all have massive effects on how it reacts in the body.
Sure they can make an ideal synthetic version of the mushroom. But that's 20 years of research to find the exact formula and then depending on its complexity another potential 5-15 years finding a way to effectively and cheaply produce it.
And sometimes the only method we can find to recreate certain compounds in manufacturing is a complicated process of additive and subtractive steps that creates byproducts we can't separate from the final product that introduce new variables to the drugs effects.
