The real reason is because they didn't know how to make her interesting so they made her the butt of everyone's jokes and a punching bag.

The in-universe reason is because the Griffin Family needs an outlet for their frustrations or they will self-destruct. She confronted them over it and when they stopped targeting her they targeted each other and the family almost destroyed itself when they started unleashing how they really felt about each other instead of using Meg as a punching bag. If I remember correctly only Lois felt remorse about it and burst into tears.
