TLDR for my prediction is Blade winning. He doesn't need the breathing gun. He's got Dracula/Hyperion scaling in base which puts him way over Buffy's physicals. So long as he gets one of his magic Zarathos level swords to parry the scythe he can simply outfight Buffy. Breathing gun is overkill and it WOULD work on Buffy. Slayers are powered by a demon and Buffy is neither holy nor pious. Cosmic Ghost Rider being bound to Zarathos was enough for the gun so Buffy being bound to an ancient shadow demon should be enough as well. I've heard the argument that because Angel's chip wouldn't let him attack Buffy due to her being human that the gun wouldn't work, but Angel's chip didn't check for human/human-demon bond, it just checked for human/non-human.
