I have found that with most women it is not what you say but what you do. Especially in the beginning non-verbal communication is infinitely more important than verbal communication.
EXAMPLES:
- Eye contact
WHEN do you look at her?
HOW you look at her?
HOW OFTEN you look at her?
FOR HOW LONG you look at her?

- Understanding of her non-verbal communication:
Do you know what her signals mean?
Can you infer her current mood from her non-verbal signals?
Can you adapt to her mood based only on non-verbal clues?
If your answer is Yes, than she will be more interested in you because she feels she doesn't have to explain herself as much for you as for someone who doesn't understand this

I can expand on the topic of non-verbal communication, especially during the early stages, but to answer your Q - There are no magic "successful" lines. You can keep some lines in your armory, but they will be more like a construction of what you need to convey in certain situation and you create lines based on these constructions

Around 30-40. I have had pretty wild 20's and it often happens that a memory of some girl that I have forgotten about suddenly appears in my mind.
Also - not all my conquests have involved full on intercourse. Some were just making out, some a bit more passionate naked making out, some were sex and some were just really nice deep conversations full of sexual tension.
So actual body count as in sex with should be around 30-40.

Really depends on how likely are you to meet that woman again. If you are colleagues and are likely to meet often for the foreseeable future, then start studying her:
Who is she friends with?
How does she dress? Does she have some personal style of just following trends?
Is she a happy person or sad?

When you get to know her periphery you will then start thinking of ways to cross her path and talk with her:
Maybe you are friends with someone who is close to her
Maybe you can arrange to have a shift together

No matter what you have spend time learning her routine at the place where you meet her, so if none of the above are optional just try to spend as much time around her and eventually an opportunity for "spontaneous conversation" will arise.

One really important thing - she should not feel like she is being stocked. You need to be extremely subtle with your information gathering. Just "happen" to be in the same place as her, but "happen" by chance doesn't mean 100% of the times. In some situations it is better to avoid going where she might be now, because you already went there yesterday and the day before.

If you have met her on the street, park, bar and you are not as likely to meet again look for something really unique about her - could be a piece of jewerly, could be her skirt, could be her make up, shoes, whatever seems unique and intriguing to you. Go to her and compliment her on this unique thing.

If you genuinely feel that this is what makes her special your "line" sound much more sincere.
