There is no solution.

Consider the grid as a 4x4 chess board. Every time you move, you switch colour. As there are 15 squares to step in, there are 15 movements, and therefore 15 colour switches. This is an odd number, so the square colour must be inverted. However the bottom right square is the same colour as the top left square, so there is a contradiction. Therefore, it is impossible.

This is just a Hamiltonian path. No? A modified DFS does the job. So assuming these are the rules:
0. you start at vertex 0 and end at vertex 15
1. you visit every vertex
2. you can visit vertices only once
Here are all the possible paths prefixed with their length. As you can see 15 vertices is the max and none include all 16.

That is the lame version. In the original you are harvesting organs and bones for billionaires in need of spare parts. In order to save money the donors only get paralyzed enough time to do the harvesting, so you don't want to enter a room with an already harvested donor before exiting the building.

I'm assuming they misworded the rules cause the person that drew it was a chink or something, because essentially the loophole to rule 2 is supposed to be that you can reenter the first room since that room didn't have a bug in it, otherwise, it would be impossible under this box arangement.

Since you havn't stepped on a bug in the starting room you can re-enter it after stepping on a bug in an adjacent room.

That isn't what the rule says at all, it doesn't say you can't enter a room with a squashed bug, it says you can't squash a bug, then re-enter a room you have already been in and you have already been in the fist room.

No, it says "After stepping on bug, no re-entering rooms", which is wrong from the original problem. The problem is impossible without being allowed to back into the starting room, because there's an even number of rooms on each side. So an Hamiltonian path that starts in opposite corners is impossible.
