For what it's worth you unlock the new missions just by starting the game and you'll unlock the new character in Chapter 3 (whenever you're meant to explore headquarters for the first time).
The new character is a scout that's almost as OP as Alicia, while the missions are a mixed bag. The default hard skirmishes feel almost like a puzzle game, because all of the enemies have stupidly overpowered interception fire (stormtroopers doing like 50 damage per shot and inflicting damage down). You can still cheese most of them with Alicia but not all of them.
I also think the DLC extra hard skirmishes are locked behind NG+ as well, but I haven't tried them because beating the game + 100%ing on the second playthrough + beating the rest of the DLC was enough content for me.
