Depending on how you look at it, Ruby might already have her ass. He canonically makes all of Sapphire's clothing and has taken her measurements in order to do so. Not only would he notice Sapphire's ass, he has to measure her whole ass to make her clothes. And since the RS/E/ORAS arcs span Sapphire undergoing puberty, Ruby would have to retake her ass measurements all the time as her body grew bigger. Ruby also just likes making clothes for her in general (their recolored Emerald arc outfits have no functional reason to exist beyond Ruby wanting them to have matching color coded outfits), so he's not restricted to only measuring her when necessary, he probably just does it for the lols too. Or if he just wants a regular excuse to grope her.
Either way, by this point Ruby probably has enough raw measurement data to plot the exponential growth of Sapphire's ass on a scientific chart. Same goes for her chest and every other part of her body. Ruby being a tailor is a great cover story to obscure the fact he's completely obsessed.
