Sure but verticality is still the main exploration force in 2D side-view games. I believe it still can work with a city built in a cliffside, with homes on different levels, winding roads going gradually down and passages going into the rocks structure. With a city like that I can have verticality, myriad secrets and all possibilities of exploration, and with in-lore explanation about people trying to live in the city with limited space I can also have freedom in designing weird, confusing paths between houses. Here's an hearly concept, with middle of the city being on a plateau and layers of buildings going down the cliffs on the side
