Yoru was usually drawn with sharper, shiftier features early on, while Asa looked softer, younger. You can't really imagine Yoru making a face like picrel. This got dropped pretty much when the weekly pace began, now they look pretty much the same.
There is also the language that Yoru uses which is very authoritative, dismissive and disrespectful of others, for most of early p2 she referred to Asa as a child.
I'm not saying it's some overt thing but she is clearly acting like a really fucked up authority figure and a bad influence on Asa, often trying to groom her into making an evil choice.
