If Earth 3 Peter gets the spider-bite then I guess he would become a corrupt TV star in league with the Government and S.H.I.E.L.D. The Maggia would be a ragtag group of resistance fighters and Black Cat would probably work with the Foreigner and other street level villains who are now heroes led by Captain Nefaria and Kingpin. Baron Zemo would probably work with Doom against Silver Sable. Symkaria is close to Latveria right? And Mary Jane as a Hobgoblin equivalent fashion designer/model could work. Or maybe her as some evil Spider themed villain working with Peter would work.
