You have to get a vocaloid editor and buy a voice bank. I used to use piapro, but then I started using vocaloid editor 6 since it's much better.
As for the Japanese I know some japanese, as for inserting lyrics. You can either manually insert lyrics into notes or use a text box that tries to insert your lyrics into the note automatically. Beware the pronounciations aren't always accurate. Sometimes something which is supposed to be pronounced as "wa" becomes "ha". Also improper pronounciations become even more of a problem with english voicebanks so I use IPA dictionary and manually check as much of the pronounciation I can manage.
Different voice banks have different tones, but within each voicebank you can adjust the tone a lot. It depends on what you do.

As for tuning vocaloids. That's a whole different beast. I think getting right pronounciations is atleast the first priority
