Hugo Strange, Renee Montoya, Harvey Bullock, Barbara Gordon, Victor Zsasz and Anarky appear. Strange is the main villain of the film. There is sequel setup involving the Court of Owls in this movie, who will be Part III's antagonists. Soundgarden's Black Hole Sun is the recurring musical motif throughout the film, in line with Nirvana's Something in the Way. No further spinoffs are in development. The Penguin Season 2 has been scrapped by DC Studios, and it's now referred to as "a limited miniseries".

No, she's just Renee Montoya, played by Eiza Gonzalez. No alter ego. Barbara Gordon briefly appears but no setup involving her being Robin or Batgirl or anything of the like. Catwoman has a recurring role but not a main role. Renee assumes that. Harvey and Gilda Dent also make brief appearances.

Anarky is a Riddler-inspired villain who Batman fights in the first and second acts and manipulated by Strange. Zsasz appears briefly. No appearance from Sofia Falcone. Penguin is barely in the script and gets defeated rather quickly by Batman and sent to Arkham.

Perhaps in response to some of the criticism surrounding Bruce Wayne in the first film, he's a lot more outgoing and playboyish in the second one.
The main plot is about Hugo Strange as Bruce's psychologist and obsessed with his Batman persona and the psychology behind it. He also turns Arkham patients into Batman-like vigilantes (like Anarky) to instigate chaos in a post-flood Gotham. He's also linked to a cabal of sorts which, obviously, turns out to be the Court of Owls but that is left for the third film to explore as Bruce uncovers their existence near Part II's end.

The only reason the sequel took this long is because of that manipulative scumfuck James Gunn trying to strongarm Reeves into quitting on it through various lowly tactics. He had the script ready for ages but Gunn kept wanting "revisions" and postponing "approving" it until Reeves just went over his head to Zazlav.

From what I know, I can only say Eiza Gonzalez has put down a deal to be in the film but other actos who have been contacted by Reeves for roles include:
1. Jared Harris as Dr. Hugo Strange (I even found a cool mock-up of him as Hugo online, kudos to whoever made this)
2. Karl Glusman as Anarky
3. Ella-Rae Smith as Barbara Gordon
4. Jesse Plemons as Harvey Dent (who COULD have an expanded role in Part III)
5. Edmund Donovan as another Arkham crazie turned deranged vigilante by Strange

Also, to address the big one, Joker does return but in a limited capacity that only functions as setup for the third film in which he will have a larger role. It seems Two-Face, The Joker will have secondary villain roles but the Court of Owls (Talon is teased) are the endgame villains for Matt Reeves' trilogy.

In the planning stages of Part II, Reeves actually considered the Affleck pitch which would've entailed Deathstroke as the villain, but he figured it was better to go his own way and found Hugo Strange to be a far more interesting villain. He also ruled it out because the character was far too "out there" for his universe. Some other scrapped plans include Black Mask, Professor Pyg and a reimagined Hush.

My source is the fact that scripts for major movies circulate like crazy and actually pass through a lot more hands than what studios would have you believe. They peddle this narrative that these scripts are lock-and-key and eyes only and all that but in reality, that's just now how it goes.

It goes through a lot of hands and, more importantly, a lot of eyes and it just so happens that one of those eyes might be a big fan (as I am) and wants to give people a taste of what the movie is about. I didn't leak the whole plot, just what it's about and what they can expect out of it. In an age where people are so invested in a movie's development only to get disappointed or bamboozled as it were, I feel like offering some transparency is good.

I also don't believe in this whole culture that's been born with social media where just saying "this movie features such and such" is some grand Watergate-level leak or something. Just putting things out there and seeing what people think as I am also a fan. I was able to read a good amount of it hence the information I divulged. I don't think any of this "ruins" the movie in any way. There's a lot of other good stuff to look forward to in it.

Zazlav himself is coming out saying what a huge success Superman is and the entire social media sphere is completely at odds with observable REALITY when the movie is actually massively flopping and will do The Marvels numbers.
