Pauline and Lady at the very least are confirmed 2 separate characters and have been for years. And Jumpman and Mario being different people is a really common explanation for there being multiple "Marios" (not just Luigi) in some of the older games.

Odyssey ending. Mario hands her a flower and she turns him down. She's only ever been extremely nice to him, but romantic feelings were never guaranteed.

So this one is complex, but there is no DKIII. Current Donkey Kong has always been a grown up Junior, but Rare got confused at one point and called Cranky DK's Grandfather. Rare also went back on that before going with it a second time and then admitting that no one actually knew wheat they were to each other and that Nintendo was responsible for deciding. And Nintendo has clearly decided that current DK is DK Jr based on the Mario Movie and MKW promotional stuff.

DK kidnapped Pauline during "March of the Minis" (a series of games), which is why I'm still not sure if Mario and DK are enemies or not. And Pauline makes note in Odyssey of being kidnapped by an ape years before and it being a traumatic ordeal. I think it's safe to assume that there is a potential for ill will between them.
