I'm not sure if you also plan on having headshots of the girls, but if you do you have a lot of work cut out for you and again, pixel art is going to be your best bet. You can hand-draw 10 basic face shapes, 10 different shoulder shapes, 10 different eye shapes, mouth shapes, nose shapes, brows, and perhaps even more hair styles, recolor them and either layer them onto each other to create permanent matches or shuffle them randomly each time using code. Given that you plan on having 256 girls, that would probably be your best option.
