You don't need to know much to start playing, but every match you should be trying to learn a little.
For starters, do bot matches they're easy enough to learn the ropes and basic mechanics in an environment where you won't just get steamrolled.
Find a hero you like, but don't pick a "main" you're going to need to find at least 3 heros you're comfortable playing with, support and tanks are easier, as they can do more with less soul farm and items, but you can play a carry(basically a DPS) if you want, you'll just need to make sure you're farming souls well in order to get your build off the ground so that you can become effective, Haze is a good character to pick if you come from a shooter background, but she is a carry and relies heavily on items to preform.
When you start a game you're in what's called a "lane phase", this lasts until you destroy their tower or they destroy your tower, you want to kill incoming creeps (the little candle npcs) and shoot their soul orb to caputre their souls, you can also shoot the enemies orange soul orbs to deny them the souls, they will do the same to you so make sure you're shooting orbs.
While doing this you also wank to poke at the enemy and get your intitial abilities set up, obviously you should take a moment in the lobby to read your abilities so you know what they do.
Once lane phase ends things get a bit more complicated, but essentially you want to rove around the map helping in team fights, collecting souls from neutral mob groups and sinners sacrifice machines(you hit them until the lights on the top start going across and you need to time your final hit so it lands when the final light is lit, this will give you some buffs), you also want to be pushing the lanes, but you can focus mostly on your original lane if you like.
Push the walkers and defend yours is the ultimate goal in this phase, eventually you will reach late game, where the shrines are being attacked, it's more of the same.
