In enhancement quests of the element you prefer. I imagine they don't drop yet for you and you're gonna have to rank up to unlock the higher difficulties that drop them. I imagine you're somewhere around Mermaid Witch which is when I'd recommend progressing through Battler mode to increase your rank and get better portraits.
You'll end up with a surplus of prism gems as you farm for the elemental stones of the megucas you're raising, but with a lack of prism stones, so I wouldn't recommend going for prism gems in the shops early on for a quick magic level increase.
Once you unlock them don't be scared of using your cubes if you're lacking in QP to raise your megucas either, you'll get more than enough.
Also, getting the free trial for the exedra passes doubles your enhancement quests drops for a month.
You'll find more info about enhancement quest materials on the 2nd link in the OP in the Upgrade & Costs tab.

At 13, you can check that in the spreadsheet.
You can go to main quests and tap/click on the rank at the top of the screen to see which rank unlocks that enhancement quest difficulty and work towards that in battler mode.

What's the general consensus on Team Damage Down vs Def Up for Defenders vs Healers? I'd imagine Defenders want Def Up and Healers want Damage Down right?

it's effectively the same whether you use def to boost defenders' barriers or hp to boost healers' heals, though you could maybe crunch the numbers to be absolutely sure. healers also usually have more trouble surviving so they could use added bulk more
or you could give them both damage reduction, since it's a stat not acquireable elsewhere. i personally prefer a mix of stat types when possible
