So from what I understand, that IS6 stage references the Battle of Hulao Pass (虎牢关), a fictional battle in Romance of The Three Kingdoms
The 3 animal dudes in that stage reference the Three Heroes, down to the preferred weapons of those people (Picrel). I'm pretty sure the arrangement of the 3 guys are also references to the order in which each of them joined in to fight Lü Bu.
As for José, I think that is a reference to Gongsun Zan (公孫瓚), who fought Lü Bu but had to retreat, prompting 1 of the Three Heroes to taunt Lü Bu into a fight. Then it became a 2v1, and then 3v1, which finally proved to be more challenging than what Lü Bu could handle.
The Battle of Hulao Pass anded with Lü Bu's defeat, though he made it out alive.

CC works by having 1 permanent map (it stays open for the entire event), and then "daily" maps (though they aren't daily anymore). Each of those stay open for a few days, and unlock new challenges to pick for the permanent map.
You can then pick and choose different challenges for the permanent map to increase your score as much as possible. Challenges mostly include stat buffs for enemies, heavy stat nerfs to your units, but there's also stuff that'll add more stage hazards, make enemies invisible, cause new/more enemies to spawn, lower your HP, gives enemies more range, etc
