I remember Misato becoming almost the main character for a bit before Kaji's death and after Asuka shows up. She has tons to do for a while and is the main person Kaji tells about his spy work.
Before Asuka, Rei seems like the second most important and Shinji and her were being set up to get together before Asuka joined and their relationship was reset.

Misato does get a lot of focus at certain points but it's mostly contained to her introduction when she starts living with Shinji to establish the dynamic, the fuckass Jet Alone episode, and then the stretch right before and after Kaji's death, with some small details about her backstory sprinkled here and there
Rei is a tougher spot since she's the main focus of the very first arc (episodes 1 through 6) with it doing a great job of presenting her relationship with Shinji going from distant coldness to mutual trust and empathy, but then she's left with very little to do until near the end because all there is to her arc is

>Shinji presents Rei with kindness and basic human decency
>this makes Rei have a change of heart
And it doesn't really come into play until very late into the plot despite it being introduced very early
