Yes disturbing the grave and reading from the book accelerated his awakening, though it's not stated directly that he would have woken up had she not read from the book, but it's kind of is implied because of all the supernatural stuff he was already controlling before she read it and him being "juicy" already rejuvenating. but the book (the other gold one) also makes him mortal at the end it's how they kill him.

If I remember correctly they don't kill him, his soul is taken to the underworld or some shit. Honestly, i never saw Imhotep as a villain. He got cursed for what was essentially a self-defense, killing a guy who would've ordered his death for the crime of touching his woman. The real villain in all of this is the system that gives men power over each other, that makes women into commodity, that allows psychopaths to torture and curse anyone who steps out of line, that pits entire nations against each other as seen in that Hamunaptra shootout, that places men like Rick on the death row and turns their deaths into entertainment for the fat slobs like the arabian jailor guy.
