Dess got lost in the dark, came into contact with the evil black glass and maybe the Wingy Dingy Man and whatever happened turned her into The Knight. The primary motivation of Carol's conspiracy is probably to turn her daughter back to normal.
I don't have to provide a complete explanation of a story that's only halfway finished, I just have to make the best case that fits with the evidence that we have.

Also, The Knight's Black Knife is made out of the "Black Shards," but that's not where they come from. The Black Shards are "black glass" growing in the depths of the Shelter.
The Black Knife came from there, it might have transformed The Knight, and Caro''s black katana was made from the shards she found in there too. Somehow, they know that the Black Shards can kill Titans, and that's what they're being prepared for.
